An interesting studie from the Medical Centre of Groningen University, Netherlands. I found a press release, no scientific paper yet.

Modern flu vaccines - incl. you seasonal flu shot - are "inactivated, split virion vaccines":

Some vaccines use an inactivated viral particle, called a split virion, which would have been broken up into tiny fragments of the original virus, each piece will contain a sample of the markers and this will allow the vaccinated individual to gain resistance to the virulent strain of the virus.

The researchers made an "old-fashioned" inactivated whole-virus vaccine, however with modern technology, to counter some disadvantages.

This resulted in a better immunity, compared to the modern inactivated split-virus vaccines.

The reason would be : a whole virus still has its nucleus with RNA; a split virus vaccine uses parts of the cell wall.

In 2006 WHO issued a report, suggesting the use of whole-virus vaccine as an option to get more vaccine available: you need a lower dose, compared to the modern vaccines.

You don't need to keep the vaccine in the fridge, room temperature is OK to store the vaccine for a year.

Also you can use freeze-drying to conserve the vaccine for a long time and for easy distribution.

This insight of better immunogenicitty of whole virus vaccine is not new; new perhaps is the way to produce and purify the vaccine. More details are awaited.

'Oud griepvaccin beter tegen pandemie'

AMSTERDAM - Het type vaccin dat in de jaren zeventig door nieuwe technologie in onbruik is geraakt werkt beter bij het bestrijden van een grieppandemie dan moderne vaccins.

Een nadeel van het oude type is dat het vooral bij kinderen nogal eens bijwerkingen veroorzaakte, maar door nieuwe technologie is van bijwerkingen waarschijnlijk geen sprake meer, concluderen onderzoekers van het UMC Groningen.

Bij de eerste griepvaccins, uit 1945, werd gebruikgemaakt van zogenoemde helevirusvaccins. Bij die methode wordt het griepvirus in grote hoeveelheden gekweekt en inactief gemaakt. Hierdoor is het virus niet meer ziekteverwekkend, maar activeert het wel het immuunsysteem.

Sinds de jaren zeventig worden vaccins geproduceerd op basis van slechts een deel van het virusmateriaal. Dit type vaccin wordt tegenwoordig gebruikt bij de jaarlijkse griepvaccinatie.

Immuunreactie
UMCG-onderzoeker Felix Geeraedts maakte vaccins van het oude type, maar dan met moderne technieken. Hij concludeerde dat die vaccins een betere immuunreactie geven dan de gangbare vaccins. Ook kunnen ze in een lagere dosis worden gegeven. Bij een grieppandemie kunnen volgens hem dan ook beter helevirusvaccins worden gebruikt. Nu is dat nog niet het geval.
De betere werking zou komen doordat bij het oude type vaccin ook materiaal uit de kern van het griepvirus aanwezig is. Juist dat kernmateriaal wekt volgens Geeraedts een betere immuunreactie op. De gangbare griepvaccins worden voornamelijk gemaakt met materiaal uit de celwand van het virus.

WHO
De onderzoeker wijst op een rapport van de Wereldgezondheidsorganisatie (WHO) uit 2006, waarin een omschakeling naar helevirusvaccins als een reële optie wordt gezien om de beschikbaarheid van griepvaccins te vergroten.
Een belangrijk extra voordeel is volgens Geeraedts dat het oude type vaccin een jaar houdbaar is op kamertemperatuur. Veel vaccins zijn alleen bij vier graden een jaar houdbaar.
