Abstract
BACKGROUND:

 The live attenuated influenza vaccine (LAIV) is the preferred vaccine for children, but the mechanisms behind protective immune responses are unclear and the duration of immunity remains to be elucidated. This study reports on the longevity of B and T-cell responses elicited by LAIV.
METHODS:

 Thirty-eight children (3-17 years old) were vaccinated with seasonal LAIV. Blood samples were collected pre-vaccination with sequential sampling up to one-year post-vaccination. Humoral responses were evaluated by hemagglutination inhibition (HI) and memory B cell (MBC) responses by ELISpot. T-cell responses were evaluated by IFN-γ ELISpot and intracellular cytokine staining for IFN-γ, IL-2, TNF-α using flow-cytometry.
RESULTS:

 LAIV induced significant increases in B and T-cell responses, which were sustained at least one-year post-vaccination. Strain variations were observed, where the B strain elicited stronger responses. IFN-γ T-cells increased significantly, and remained higher than pre-vaccination levels one year later. Th1 intracellular cytokines (IL-2, IFN-γ, TNF-α) increased after one dose and were boosted post-second dose. HI titers were sustained for one year. Vaccine induced MBCs were significantly increased and the response persisted 360 days later.
CONCLUSION:

 LAIV vaccine elicited B and T-cellular responses that persisted at least one year in children, a novel finding aiding future vaccine policy.
